Reconstitution and storage guide

Bench procedures for handling lyophilized research peptides: reconstituting, calculating concentration, and storing the result.

Reconstituting a lyophilized vial

  1. Let the vial reach room temperature before opening to avoid condensation on the powder.
  2. Swab the stopper with an alcohol prep pad and let it dry.
  3. Draw the chosen volume of bacteriostatic water into a sterile syringe.
  4. Insert the needle and let the solvent run slowly down the inside wall of the vial, not directly onto the powder.
  5. Swirl gently until the solution is clear. Never shake; shear can fragment long peptides.
  6. Label the vial with the date of reconstitution and the final concentration.

Working out concentration

Concentration equals vial content divided by solvent volume. A 10 mg vial reconstituted with 2 mL gives 5 mg/mL, or 5,000 µg/mL. Use the calculator at the side to check your numbers.

Vial contentSolvent addedConcentrationµg per 0.1 mL

Storage

Lyophilized, sealed−20 °C, dark. Stable for 24 months or more for most peptides.
Lyophilized, short term2–8 °C for up to 30 days, for example in transit or on a shared bench fridge.
Reconstituted2–8 °C, use within 7 days unless the product page states otherwise. Do not freeze reconstituted solutions.
Bacteriostatic waterRoom temperature. Discard 28 days after first puncture.

Choosing a solvent

Bacteriostatic water suits most peptides and allows repeated withdrawal from the same vial. Use sterile water without preservative for compounds that react with benzyl alcohol. A few hydrophobic peptides need a small amount of acetic acid or DMSO before dilution; the product page notes this where it applies.
